Day e filloi karrieren e saj si kengetare e njohur ne vitin 1939. Popullariteti i saj filloi te rritet pas regjistrimit te pare te saj me hitin , "sentimentale Journey", ne vitin 1945. Pas largimit Les Brown & Band provoi nje karriere solo, ajo filloi nje partneritet te qendrueshem me Columbia Records, kenge te cilat do te mbeten i vetmi regjistrim i etiketuar. Kontrata zgjati 1947-1967, dhe ka perfshire me shume se 650 incizime, duke e bere Day-n nje nga kengetaret me te popullarizuara dhe te vleresuara te shekullit te 20. Ne vitin 1948, pasi u bind nga Sammy Cahn, Jule Styne ne ate kohe dhe agjenti Al Levy, ajo auditoi per Michael Curtiz, qe e coi ate ne rolin "Romance ne det te hapur".
Gjate karrieres se saj, Day u shfaq ne 39 filma. Ajo u rendit si ylli me i mire box office, gruaja e vetme ne ate liste, per kater vjet (1960, 1962, 1963 dhe 1964)ne rangun 10 me te larte per dhjete vjet (1951-1952 dhe 1959-1966). Ajo u be te rangut te larte femer yll kuti-zyra e te gjitha koherave dhe aktualisht renditet e gjashta nder 10 interpretues me te larte (meshkuj dhe femra), qe nga 2012. Ajo u propozua per Academy Award per rolin e saj ne Pillow Talk, fitoi tre Henrietta Awards (World Film Favorite), mori Los Angeles Film Critics Association's Career Achievement Award, ne vitin 1989, mori cmimin Cecil B. DeMille per arritje jetesore ne filma. Doris e realizoi filmin e saj te fundit ne vitin 1968.
